Magnesium glycinate is sold everywhere as the “best absorbed” form of magnesium. The problem: the official NIH list of well-absorbed magnesium forms doesn’t actually include it. Here’s what the research does and doesn’t say about absorption, sleep, and how much is actually safe to take.

How Magnesium Absorption Actually Works — and What “Better Absorbed” Really Means
Most of the magnesium in your body sits in bone and soft tissue, and your kidneys tightly regulate how much stays in your blood. When you eat magnesium-rich food, your body typically absorbs about 30 to 40 percent of it. Supplements are a different story — absorption varies a lot by form, largely based on how well that form dissolves in liquid.
According to the NIH Office of Dietary Supplements, small studies have found that magnesium in aspartate, citrate, lactate, and chloride forms is absorbed more completely than magnesium oxide and magnesium sulfate. That’s the closest thing to an official, agency-backed ranking of magnesium forms that exists. Glycinate isn’t on that list — not because it’s been tested and found lacking, but because there simply isn’t enough large-scale research in healthy adults to include it one way or the other.
So where does the “best absorbed” claim about glycinate actually come from?
The Actual Evidence Behind the Glycinate Absorption Claim
The most direct comparison of magnesium glycinate’s absorption comes from a 1994 clinical trial published in the Journal of Parenteral and Enteral Nutrition. Researchers gave 12 patients who’d had part of their small intestine surgically removed — a condition that impairs mineral absorption — a radioactively labeled dose of either magnesium diglycinate (chelate) or magnesium oxide, then tracked how much was actually absorbed.
Across the whole group, there was no significant difference: 23.5 percent absorption for the glycinate form versus 22.8 percent for magnesium oxide. The difference only showed up in the four patients with the worst underlying absorption problems, where glycinate outperformed oxide by a wide margin (23.5 percent versus 11.8 percent). It’s a real finding, but it comes from a small group of patients with impaired intestines, not from healthy adults — which makes it a shaky foundation for a blanket “glycinate absorbs best” claim.
What the same research does support is tolerability. Magnesium oxide, carbonate, chloride, and gluconate are the forms most consistently linked to diarrhea, largely because unabsorbed magnesium in the gut pulls in water. Glycinate isn’t on that list either. A product like Doctor’s Best’s magnesium glycinate lysinate is a reasonable pick less because it’s guaranteed to out-absorb everything else, and more because it’s less likely to send you to the bathroom at the doses people typically take.
Absorption is one question. Whether taking it actually changes how you sleep or feel is another.
Does It Actually Help You Sleep or Feel Less Anxious?
On sleep, a 2023 systematic review pooling 9 studies and more than 7,500 people found that observational studies showed an association between magnesium status and sleep quality — people with lower magnesium tended to report worse sleep. But when researchers actually tested magnesium supplementation in randomized trials, the results were inconsistent enough that the authors called the relationship “uncertain.”
The most recent and directly relevant trial is a 2025 randomized, placebo-controlled study that gave 155 adults with self-reported poor sleep 250mg of elemental magnesium a day, in the bisglycinate form, for four weeks. Insomnia severity scores dropped more in the magnesium group than the placebo group (a difference of about 1.6 points), but the result was only barely statistically significant (p = 0.049), and the researchers themselves described the effect size as “small.” One detail stood out: people who started with lower dietary magnesium intake saw bigger improvements — the same pattern of “more benefit if you were actually low to begin with” that shows up across most magnesium research.
Anxiety tells a similar story. A 2017 systematic review of 18 studies found that magnesium reduces neuronal excitability partly by inhibiting NMDA receptor activity and supporting GABA signaling — the same inhibitory system targeted by anti-anxiety medications. But only about a third of the reviewed studies in mildly anxious people showed a real benefit, and most of those combined magnesium with vitamin B6, making it impossible to say how much magnesium alone contributed. The review’s authors were blunt about it: “the quality of the existing evidence is poor.” It’s also worth noting that none of the 18 studies in that review tested glycinate specifically — most used lactate, oxide, or citrate — so a lot of the popular “magnesium glycinate for anxiety” claims are really borrowing evidence from other forms of magnesium entirely.
None of this means magnesium is useless — it just means the size of the benefit, and who’s likely to notice it, matters more than the marketing suggests.
How Much Should You Actually Take?
The recommended dietary allowance for magnesium is 400 to 420mg a day for adult men and 310 to 320mg for adult women, counting food and supplements combined. Despite that, an analysis of national nutrition survey data found that 48 percent of Americans consume less magnesium than their estimated average requirement from food and drinks alone — so a real gap between recommended and actual intake exists for a lot of people.
There’s also an upper limit worth knowing. The NIH sets the tolerable upper intake level for supplemental magnesium (meaning from pills or medication, not food) at 350mg a day for adults. This particular product’s label directs adults to take two tablets twice a day, which works out to 400mg of elemental magnesium daily if taken at the full labeled amount — above that 350mg supplemental limit. That’s not a red-flag-level risk for most healthy adults; the main consequence of going over is a higher chance of loose stools, not the kind of serious toxicity that shows up at much higher doses (typically several grams a day, usually from laxatives or antacids). Still, it’s the kind of detail worth checking against the label rather than assuming the full recommended dose is automatically the right one for you.
A few other things worth knowing, per the NIH: magnesium can reduce the absorption of oral bisphosphonates used for osteoporosis, and it can bind with tetracycline and quinolone antibiotics, so those are typically spaced at least two hours apart from a magnesium dose. Long-term use of proton pump inhibitors can also lower magnesium levels in the body, which is a separate issue from how much magnesium you take in supplement form.
The Bottom Line
“Best absorbed” isn’t a claim the NIH’s own list of well-studied magnesium forms actually backs up for glycinate — the direct evidence comes from a small study in patients with impaired intestines, not healthy adults. What does hold up is that glycinate tends to be gentler on the stomach than forms like oxide or carbonate. As for sleep and anxiety, the research suggests a real but modest effect, more noticeable in people who were already running low on magnesium than in people who weren’t. And if you’re taking this specific product at its full labeled dose, it’s worth knowing that adds up to slightly more than the official upper limit for supplemental magnesium.
That fancier price tag isn’t buying you a spot on the NIH’s list of best-absorbed forms — but it might be buying you an easier time on your stomach, and that’s worth something too.
